Transient ischemia/reperfusion has been shown to evoke increases in enkephalin levels in myocardial tissue. Blockade of δ opioid receptors eliminates the cardioprotective effect of ischemic preconditioning both in vivo and in vitro. Inhibition of κ1 opioid receptors eliminates the cardioprotective effect of preconditioning only in vitro. Agonists of μ, δ1, δ2, and κ1 opioid receptors imitate the cardioprotective effect of preconditioning. These results indicate that endogenous opioid peptides are triggers of ischemic preconditioning.
