Most of the CARs employ single-chain fragment of antibodies (scFv) as a target recognition module. ScFv, however, has some weak points, namely two-domain format and almost 30kDa weight. Here we describe CAR with the 10th human fibronectin type III domain as alternative antigen recognition module of CAR. We designed and developed mono- and bispecific Fn3CARs as a recognition module and proved their functionality.
